Natural killer (NK) cells are immune cells that can kill cancer cells without needing a specific antigen match, making them promising for 'off-the-shelf' therapies. However, treating solid tumors with NK cell immunotherapy is much harder than treating blood cancers. The main challenges include getting NK cells to enter the tumor, surviving the hostile environment inside the tumor, and finding targets that are present on all cancer cells.

A key challenge is the immunosuppressive tumor microenvironment (TME). Solid tumors create a hostile environment that exhausts immune cells, including NK cells, and limits their ability to function 39.
