Key Takeaway
Recognize m6A-related enzymes as potential tools for disease prediction and monitoring allergen immunotherapy efficacy.

This narrative review examines the role of m6A modification in the pathogenesis of allergic airway diseases. The authors synthesize evidence regarding how the dysregulation of m6A modification contributes to several key pathological processes, including the disruption of epithelial barriers, the induction of oxidative stress, and the activation of inflammatory cells. These processes are linked to the remodeling of airways in patients with these conditions.

Furthermore, the review discusses m6A-related enzymes as potential clinical tools. These enzymes may be utilized for disease prediction, as targets for therapeutic intervention, and as markers to evaluate the efficacy of allergen immunotherapy.

While the review identifies these mechanisms as significant, the evidence regarding m6A-targeted therapeutic strategies is currently preliminary. The clinical utility of m6A-related enzymes for predicting disease or monitoring immunotherapy remains a subject for further investigation. The scope of the review is focused on the molecular mechanisms and potential diagnostic/therapeutic applications of m6A modification in the context of allergic airway diseases.

Living with allergic airway diseases can mean dealing with constant inflammation and damaged airways. Researchers are looking closely at a specific process called m6A modification. This is a chemical change that happens to cells, and when it goes wrong, it can cause serious problems for the body's defenses.

The review shows that when m6A modification is out of balance, it leads to several issues. These include breaking down the protective barrier of the airway, causing oxidative stress, and triggering inflammatory cells. These factors together can lead to airway remodeling, which is the physical changing of the airway structure.

While we are still learning, these m6A-related enzymes could become useful tools in the future. They might help doctors predict disease, create new treatments, and even check how well allergy shots are working. Because this is a narrative review, these findings are early steps toward understanding how to better manage these conditions.

What this means for you:
M6A modification is a key driver of inflammation and airway damage in allergic diseases.

Common questions

What is m6A modification and why does it matter?

M6A modification is a chemical change in cells. When this process is out of balance, it can cause significant problems like breaking down the protective barrier of the airway, causing oxidative stress, and activating inflammatory cells. This can lead to the remodeling of airways in people with allergic diseases.

How can these findings help people with allergies?

M6A-related enzymes could eventually be used as tools for several purposes. They may help doctors predict disease, create new ways to treat the condition, and evaluate how well allergen immunotherapy, such as allergy shots, is working for a patient.

N6-methyladenosine (m6A) represents the most prevalent modification on eukaryotic RNA and plays a critical role in nearly all stages of RNA expression regulation. Extensive research demonstrates that the dysregulation of m6A modification plays a significant role in the disruption of epithelial barriers, the induction of oxidative stress, the activation of inflammatory cells, and the remodeling of airways, all of which are characteristic pathological features of allergic airway diseases (AADs). Given the distinct roles of various cell types in the progression and resolution of AADs, understanding how m6A functions across cellular contexts is essential for a comprehensive understanding of the disease’s pathological mechanisms. This review begins by outlining the fundamental biological characteristics of m6A modification. Subsequently, we explore how dynamic m6A modifications affect AADs progression from the perspectives of airway epithelial cells, innate immune cells, and adaptive immune cells, emphasizing the pivotal regulatory role of m6A in epithelial-immune crosstalk. We then summarize how cellular context shapes the functional heterogeneity of m6A modification and discuss the potential underlying mechanisms. Finally, we summarize the latest progress in the use of m6A-related enzymes as tools for disease prediction, therapeutic intervention, and evaluation of allergen immunotherapy efficacy, and discuss the opportunities and challenges associated with the clinical translation of m6A-targeted therapeutic strategies.
