Anal Cancer: what the trials found

In the management of anal cancer and related conditions, carboplatin has demonstrated a statistically significant improvement in progression-free survival (PFS) compared to alternative treatments (9.3 vs. 7.4; p=0.0006) 5. Clinical observation was also utilized to monitor high-grade anal intraepithelial neoplasia at one year 4.

Immunological and prophylactic interventions show significant results in HPV management. V503 significantly reduced the combined incidence of HPV 6/11/16/18-related anogenital persistent infection (0.2 vs. 2.2; p=<0.001) 6. The 9vHPV vaccine demonstrated significant differences in geometric mean titers (GMTs) across various HPV types (p=<0.001) 7.

Treatment with imiquimod resulted in a reduction in physical symptom scores from baseline to four weeks post-randomization (-2.22 vs. -0.08) 3. Regarding cetuximab, trials showed no statistically significant difference in local failure rates at three years (0.259 vs. 0.353; p=0.218) 19, while another study reported locoregional failure rates of 42 and 20 respectively with a recorded QoL EORTC Global Score of 78.9 at one year 20.

Fludarabine was evaluated in clinical settings [17, 18].

Recent results — preliminary, needs further review

  • Acupuncture therapy was evaluated for chemotherapy-induced peripheral neuropathy (CIPN); results showed a mean change in severity of 5.9 and a reduction in the number of participants with Grade 2 or higher CIPN to 3 1.
  • Non-interventional data is available for a larger cohort of 460 patients 2.

For the clinician treating this condition

  • Carboplatin is associated with statistically significant improvements in progression-free survival (PFS) 5.
  • V503 and the 9vHPV vaccine are established interventions for managing HPV-related infections and titers, respectively [6, 7].
  • Imiquimod has been shown to reduce physical symptom scores in patients 3.
