Bone metastases: what the trials found

Several trials evaluated the efficacy of Denosumab in managing skeletal-related events (SREs). Studies demonstrated significant improvements in time to first SRE, with results showing statistically significant non-inferiority and superiority across different trial cohorts [1, 6, 8].

Zoledronic acid was shown to significantly improve the time to the first SRE, demonstrating both non-inferiority and superiority in clinical outcomes 7.

Radium-223 dichloride (Xofigo) demonstrated significant improvements in overall survival, time to total alkaline phosphatase (ALP) progression, and percentage of patients with a total ALP response at week 12 5. Safety data for Radium-223 also established the frequency of high-grade treatment-emergent adverse events 13.

Hypofractionated radiation treatment was associated with significant improvements in pain response among evaluable participants 2.

Recent results — preliminary, needs further review

  • A study on CT-guided bone percutaneous procedures is currently under investigation and not yet corroborated 16.

For the clinician treating this condition

  • Denosumab and Zoledronic acid are established interventions for reducing the time to first skeletal-related events (SREs) in patients with bone metastases.
  • Radium-223 dichloride is an established treatment associated with improved overall survival and progression-free metrics in specific patient populations [5, 13].
  • Hypofractionated radiation therapy has been shown to improve pain response in patients with bone involvement 2.
