Crohn disease: what the trials found

Several biologics have demonstrated efficacy in Crohn disease. Vedolizumab showed clinical response rates of 72.9% at week 14, 70.8% at week 30, and 68.8% at week 46 5. In a separate study, Vedolizumab achieved endoscopic remission in 34.5% of participants and endoscopic healing in 43.6% of participants at week 26 2.

Ustekinumab showed varying results across trials. One study reported clinical response rates of 49.1% and 37.4% at week 16 12, while another reported endoscopic response rates of 37.9% and 29.9% at week 48 8. A placebo-controlled trial for Ustekinumab showed clinical response rates of 66.2% and 72.3% at week 52 7. A separate study of Ustekinumab showed no significant difference in steroid-free clinical remission or endoscopic remission at week 48 1.

Other established treatments include infliximab, which showed significantly higher rates of corticosteroid-free clinical remission (up to 72.2%) and mucosal healing (up to 43.9%) in a multi-arm study 16. Certolizumab pegol showed clinical response in 40.5% and 34.0% of patients at week 6 15. BI 695501 demonstrated high clinical response rates at week 4 (88.0% and 93.1%) and week 24 (87.4% and 87.4%) 10.

Ozanimod showed no significant difference in CDAI scores, abdominal pain/stool frequency remission, or SES-CD score decreases compared to its comparator 11. Etrolizumab showed varying clinical remission rates at week 14 across different cohorts 9. In a study of RHB-104, patients on the combination showed a significantly shorter duration of time in remission compared to the comparator 13.
