Neoplasms: what the trials found

Several agents have been evaluated for the management of neoplasms, with varying outcomes in efficacy and safety. Bevacizumab showed adverse event rates ranging from 71.4% to 100.0% 5. Everolimus demonstrated clinical benefit in approximately 7% to 9% of patients across different cohorts, with associated adverse events reported at 1% to 9% 6. Vemurafenib was associated with adverse or serious adverse events in 19.9% to 93.2% of participants depending on the cohort 7.

In specific clinical contexts, Sorafenib showed a range of severe treatment-related adverse events (CTCAE Grade 3 or higher) across different arms 13. Nab-paclitaxel demonstrated a statistically significant improvement in Overall Survival (OS) with p=0.0128 10, while the reported number of high-grade laboratory abnormalities was low across groups 10. Cabazitaxel showed statistically significant improvements in Overall Survival (p<0.0001), Progression Free Survival (p<0.0001), and Overall Tumor Response (p=0.0005) 16.

Other treatments included Herceptin, where zero participants experienced a drop in Left Ventricular Ejection Fraction below 45% 12, and Rivaroxaban, which showed no statistically significant difference in the time to first occurrence of major bleeding events (p=0.265) or primary efficacy endpoints (p=0.101) 9. For pain management in oncology settings, Tapentadol ER resulted in 84% to 98% of patients achieving pain control 15, while Botulinum Toxin Type A did not show a statistically significant difference in average pain scores compared to the control (p=0.43) 14.

Recent results — preliminary, needs further review

  • BI 754091 showed no dose-limiting toxicities in Phase Ia, but reported adverse events in 96.3% to 100% of participants and serious adverse events in 25.9% to 48.4% of patients during Phase Ib 2.
  • Ezabenlimab demonstrated disease control rates ranging from 38.9% to 86.7% across different cohorts 3.
  • CC-90011 was associated with varying numbers of high-grade hematology and chemistry toxicities, as well as post-baseline ECOG status shifts 21.
  • Trastuzumab showed disease control rates ranging from 16.2% to 66.7% across various tumor-pathway cohorts 22.
  • Dostarlimab was evaluated for hematology and clinical chemistry results, with varying counts of high-grade increases across arms 17.

For the clinician treating this condition

  • Cabazitaxel demonstrated statistically significant improvements in OS, PFS, and overall tumor response compared to the control 16.
  • Nab-paclitaxel showed a statistically significant improvement in Overall Survival (p=0.0128) 10.
  • Treatment outcomes for newer agents like Ezabenlimab and Trastuzumab show varying rates of disease control across different patient cohorts [3, 22].
