Adenocarcinoma: what the trials found

Combination therapy with Gemcitabine and Cisplatin demonstrated significant improvements in clinical outcomes, including progression-free survival (11.01 vs. 5.59; p<0.0001), objective response rates (67.8% vs. 23.0%; p<0.0001), and disease control (92.6% vs. 76.2%; p<0.0001) 4. Gemcitabine monotherapy also showed evidence of anti-tumor activity, with a 28% proportion of patients remaining free of disease progression at six months compared to 14% in the control group 19, and a clinical benefit response rate of 33% vs. 0% 19.

Pemetrexed showed significant improvements in progression-free survival (11.17 vs. 6.90; p=0.0002), objective response rates (56.5% vs. 22.6%; p<0.0001), and disease control (90.4% vs. 80.9%; p=0.0118) 6. Ramucirumab was associated with improved overall survival (5.2 vs. 3.8; p=0.0473) and progression-free survival (2.1 vs. 1.3; p<0.0001), with a significantly higher percentage of patients remaining progression-free at week 12 (40.1% vs. 15.8%; p<0.0001) 5.

Nab-paclitaxel was evaluated in multiple trials [2, 11]. In one study, the incidence of peripheral neuropathy or myelosuppression did not differ significantly between groups (76.5% vs. 77.1%; p=0.9258), while dose reductions occurred in approximately 57-65% of participants across treatment arms 2. Bevacizumab was also evaluated in a large cohort, though specific outcome metrics for that trial were not provided [7, 22].
