Extensive Stage Small Cell Lung Carcinoma: what the trials found

Carboplatin has been evaluated in several trials for the management of Extensive Stage Small Cell Lung Carcinoma. Results regarding progression-free survival (PFS) were mixed; one study showed a statistically significant improvement in PFS with Carboplatin (3.7 vs 2.1, p=0.02) 9, while another trial did not find a statistically significant difference in PFS (5.5 vs 4.9, p=0.17) 4.

Cisplatin demonstrated varying results across studies. One study reported a statistically significant improvement in progression-free survival (p<0.001) 7, while another trial showed no significant difference in PFS between groups (p=0.45) 8. Additionally, one Cisplatin trial noted a non-significant difference in overall survival (OS) (10.3 vs 8.9, p=0.17) 7.

New data regarding Carboplatin has been established in Phase 1 trials 1.

Recent results — preliminary, needs further review

  • Auranofin was evaluated in a Phase 2 trial where 2 out of 3 patients experienced Grade 3+ adverse events 3.

For the clinician treating this condition

  • Evidence regarding Carboplatin for progression-free survival is inconsistent across trials, with one study showing significant improvement and another showing no significant difference.
  • Cisplatin results are also mixed; while one trial showed a statistically significant improvement in progression-free survival, another did not reach significance.
  • Safety data for Auranofin indicates a low incidence of Grade 3+ adverse events in early phase testing.
