Mumps: what the trials found

Several phase 3 trials demonstrate high seropositivity rates for mumps following vaccination with various MMR-containing vaccines. For instance, Priorix showed anti-mumps antibody concentrations above the cut-off in 98.0-99.7% of subjects across multiple studies 57. Similarly, ProQuad achieved mumps seropositivity rates of 96.3-99.5% after dose 2 489. A phase 4 trial of the SA 14-14-2 Japanese Encephalitis vaccine reported mumps seropositivity of 98.5% 1.

Concomitant administration of MMR with other vaccines (e.g., Japanese encephalitis chimeric virus vaccine) did not impair mumps seroconversion, with rates of 96.9-100% 13. However, one trial comparing MMRV to MMR+V found lower mumps antibody levels (≥10 Units/mL) in the MMRV group (97.2% vs 98.2%, p<0.001) 14, indicating mixed evidence for combined vaccines.

Safety profiles were generally acceptable. Solicited injection-site adverse reactions after ProQuad dose 2 occurred in 10.4-17.2% of participants 4.
