Neuroendocrine Tumor: what the trials found

Axitinib demonstrated a statistically significant improvement in Objective Response Rate (ORR) compared to the control group (p=0.007) 1. However, Axitinib did not show statistically significant differences in Progression-Free Survival (PFS) 1 or Biochemical Response 1.

Everolimus demonstrated a statistically significant improvement in the probability of participants remaining event-free in PFS based on central radiology assessment (p=<0.001) 6. Other studies evaluating Everolimus showed a 47% rate of hepatic progression-free survival at 24 months 20, though other metrics such as overall survival and first/second progression-free survival did not reach statistical significance in those specific trials [6, 7].

Nivolumab was evaluated in two studies; one reported a 54.1% overall survival rate at 12 months 17. Other established interventions include Octreotide (including long-acting release) [3, 10], Pasireotide 4, and various imaging agents such as 64Cu-DOTATATE 8 and 68Ga-DOTATOC 9.

RAD001 showed a 44.9% ORR and a 73.5% disease control rate 2.

Recent results — preliminary, needs further review

  • Lenvatinib was evaluated in a Phase 2 trial 11.

For the clinician treating this condition

  • Axitinib provides a statistically significant improvement in Objective Response Rate (ORR) but does not significantly impact PFS or biochemical response 1.
  • Everolimus shows significant improvement in progression-free survival based on central radiology assessment 6 and maintains hepatic progression-free survival at 24 months 20.
  • Nivolumab has been established in clinical trials with a reported 12-month overall survival rate of 54.1% 17.
