What the trials found Updated — new results For clinicians

Sarcoma: what the trials found

Established evidence for sarcoma treatment includes the use of dactinomycin, which showed a 36% event-free rate at 4 years following study entry; however, concurrent therapy was associated with adverse events in 53.3% to 79.3% of patients 6. Trabectedin demonstrated objective response rates ranging from 5.9% to 27.0% 4.

Pharmacological studies on other agents show that glutamic acid administration resulted in recorded neurotoxicity at various intervals 5. Additionally, the incidence of toxicity related to VI treatment in patients with UGT1A1 genotypes was not significantly different across groups (p=0.99) when administered with Cyclophosphamide 3. Sodium thiosulfate has also been evaluated in established clinical trials for this condition 2.

Recent results — preliminary, needs further review

  • Pembrolizumab is currently being evaluated in a Phase 2 trial 1.
  • Eribulin is under investigation in a Phase 2 trial 8.
  • A combination of MK3475 with Metronomic CP showed no objective response at 6 months, though non-progression rates varied between 0.0% and 40% across study arms; however, no patients remained alive with a best overall response 7.

For the clinician treating this condition

  • Dactinomycin is associated with significant adverse events when used in concurrent therapy (up to 79.3%) but maintains a 36% event-free rate at 4 years 6.
  • Trabectedin shows variable objective response rates ranging from 5.9% to 27.0% 4.
  • Cyclophosphamide treatment and UGT1A1 genotype do not appear to significantly correlate with the incidence of toxicity related to VI treatment (p=0.99) 3.
