Solid malignancies: what the trials found

Clinical evidence for solid malignancies includes the use of MEDI4736 (Durvalumab) in a Phase 3 trial involving 867 participants 1.

Pharmacodynamic and safety evaluations were conducted for MLN0128 in two separate Phase 1 trials. In one cohort of 198 patients, MLN0128 demonstrated significant reductions in p4EBP1 (-64.1%), pAKT (-15.5%), and PS6 (-99.5%) from baseline 3. In a smaller cohort of 68 patients, the treatment was associated with a 100% incidence rate for various adverse event categories, including serious adverse events and those resulting in discontinuation, while 63% of participants experienced fatal adverse events within 30 days of the last dose 4.
