Cachexia: what the trials found

Anamorelin HCl has demonstrated significant improvements in lean body mass and A/CS domain scores in patients with cachexia [1, 2]. Specifically, trials showed a change in lean body mass of 0.99 and 0.65 respectively (p<0.0001) and A/CS domain score improvements of 4.12 and 3.48 (p=0.0004; p=0.0016) [1, 2]. One study also reported a change in A/CS domain scores of 4.46 vs 3.24 3.

In early-phase studies for PF-06946860, patients showed changes in appetite scores at week 4 (2.45; 1.84) and across multiple weeks (ranging from 0.99 to 2.30) 11. Fatigue scores were also recorded over a 6-week period following administration 11.

Recent results — preliminary, needs further review

  • * Ponsegromab showed no significant change in physical activity or mean activity level during maximum 6 minutes at week 12 (p=0.8260; p=0.5762) 4. * SUN11031 was associated with changes in lean body mass of 37.85 to 40.39 across various time points in patients with COPD-associated cachexia 5. * BYM338 showed a 4.97% increase in total lean body mass (LBM) via DXA at week 8 compared to 2.41% for placebo 8. * Placebo Testosterone was associated with changes in total lean body mass of -3.31 vs 1.42 and improvements in peak isometric leg strength 13. * In-person nutritional counseling by a registered dietitian resulted in estimated mean weight changes of -0.1, -0.2, and -1.2 over 6 months 14.

For the clinician treating this condition

  • Anamorelin HCl is associated with statistically significant increases in lean body mass and improvements in A/CS domain scores [1, 2].
  • PF-06946860 has been shown to impact appetite scores in patients with cancer-related cachexia 11.
