Prurigo nodularis: what the trials found

Nemolizumab has demonstrated significant efficacy in improving pruritus and clinical outcomes. In two Phase 3 trials, patients receiving Nemolizumab showed significantly higher rates of achieving a ≥ 4-point improvement on the Peak Pruritus Numeric Rating Scale (PP NRS) at both Week 4 and Week 16 compared to placebo [3, 4]. Additionally, Nemolizumab was associated with significantly higher Investigator Global Assessment (IGA) success rates at Week 16 [3, 4]. Long-term data showed high rates of maintaining IGA success through scheduled visits 8.

Nalbuphine ER tablets demonstrated statistically significant improvements in itch intensity and quality of life. Patients treated with Nalbuphine showed a significantly higher percentage of ≥ 4-point decreases in the Worst Itch Numeric Rating Scale (WI-NRS) at Week 14 7. This was accompanied by significant improvements in the Itch-related Quality of Life (ItchyQoL) score and the Prurigo Activity Score (PAS) 7.

Serlopitant trials showed mixed results regarding responder rates. In one study, there was no statistically significant difference between Serlopitane and placebo for WI-NRS 4-point responder rates at Week 10 9. However, in another trial, a significantly higher percentage of patients achieved a WI-NRS 4-point response at Week 4 when treated with 5mg Serlopitant compared to placebo 11.

Other interventions include the use of corticosteroid with lidocaine 2, nalbuphine HCl ER tablets [5, 10], and Ruxolitinib Cream. However, Ruxolitinib Cream did not show statistically significant improvements in WI-NRS4 response at Week 4 or Week 12, nor did it show a significant difference in overall treatment success at Week 12 6.
