New evidence indicates that Ataluren showed a statistically significant reduction in 6-Minute Walk Distance (6MWD) at week 72 when analyzed using an Intent-to-Treat (ITT) population 1. However, this same metric did not reach statistical significance in the Modified Intention-to-Treat (mITT) population 1. In a separate study of Ataluren, no significant change was observed in 6MWD at week 48 2.

Regarding agitation management, Brexpiprazole combined with OPC-34712 demonstrated a statistically significant reduction in the Cohen-Mansfield Agitation Inventory (CMAI) total score after 12 weeks of treatment 5, though it did not significantly impact Clinical Global Impression-Severity of Illness (CGI-S) scores related to agitation 5. A different trial using the same combination did not reach statistical significance for CMAI scores 3.

In the management of seizures, SHP615 demonstrated a response rate of 80% in a small cohort 8. In this study, patients showed sustained absence of seizure activity for at least 1, 4, and 6 hours at rates of 68.0%, 36.0%, and 32.0%, respectively 8. A smaller trial reported that no participants required additional anticonvulsant medication for ongoing status epilepticus after administration of SHP615 7.

For fibromyalgia, TNX-102 SL (2.8mg) showed a reduction in both the Fibromyalgia Impact Questionnaire - Revised (FIQR) Symptoms Domain Score and Function Domain Score at week 12 6.
