Non-Alcoholic Fatty Liver Disease: what the trials found

Diagnostic imaging research has established correlations between MRI-PDFF liver fat percentages and various ultrasound biomarkers. Specifically, MRI-PDFF measurements showed a correlation of 1.7 with the Hepatorenal Index (HRI) [4, 5], a correlation of 0.6 to 1.0 with Acoustic Attenuation [4, 5], and a correlation of 5.9 to 6.6 with Tissue Stiffness [4, 5].

Pharmacological interventions have been evaluated in several phases. Pioglitazone was studied for its impact on histological scores and the prevention/reversal of T2DM or pre-diabetic states 6. Other established trials included evaluations of OMACOR 7, Exenatide [8, 9], Semaglutide 10, and SAMe 11.

Recent results — preliminary, needs further review

  • Vitamin E demonstrated a significant reduction in serum Alanine Aminotransferase (ALT) from baseline to 24 weeks (p=<0.0001), with approximately 10-24% of patients achieving ALT normalization (p=0.004) 2.
  • Efinopegdutide was associated with mean body weight reductions at week 28 (-3.9% to -7.5%) and significant reductions in liver fat content (LFC) via MRI-PDFF at 24 weeks (p=<0.0001) [1, 18].
