Plaque psoriasis: what the trials found

Several biologics demonstrate significant efficacy in treating plaque psoriasis. Bimekizumab achieved a PASI90 response rate of 94.0% and an IGA 0/1 response rate of 92.0% at week 16 1. Ixekizumab demonstrated high rates of PASI 90 (58.5%) and PASI 75 (22.9%) in a large-scale study, with significant superiority over the comparator in both metrics 7. Additionally, Ixekizumab showed an 86.0% PASI-75 response rate at week 12 in patients with no active psoriatic arthritis 13.

JNJ-77242113 demonstrated consistent improvements across multiple trials; notably, it achieved a PASI-75 of 58.1% to 78.6% and PASI-90 rates ranging from 25.6% to 59.5% at week 16 [3, 5]. Secukinumab showed significant PASI 90 results at week 52 (70 participants vs 32 in the comparator) 14, and was associated with high DLQI 0/1 response rates of approximately 61.7% to 74.7% at both weeks 16 and 52 9. Tildrakizumab showed sustained PASI scores through week 96 6.

Etanercept achieved a PASI-75 response of 41.6% by week 12, with PASI-50 rates reaching up to 74.0% during the observation period 11. Apremilast demonstrated significant improvements in nail psoriasis, including a mean 64.1% reduction in mNAPSI at week 36 and a 50.42% reduction in patient-reported nail pain by week 52 19.

Various topical and localized treatments were evaluated: DFD-01 (betamethasone dipropionate) spray resulted in reductions in Investigator's Global Assessment grade (-0.8) and body surface area involvement (-1.3) 21. Lac-Hydrin lotion combined with UItravate ointment showed significant improvements in physical global assessment and signs of psoriasis, atrophy, or telangiectasis 12.
