Postmenopausal Osteoporosis: what the trials found

New clinical evidence identifies ENZ215 as a treatment for postmenopausal osteoporosis 1.

Teriparatide significantly reduces the incidence of new vertebral fractures (p=0.000094), pooled new and worsening vertebral fractures (p=0.000075), and pooled clinical vertebral and non-vertebral fragility fractures (p=0.000869) 2.

Denosumab is associated with significant increases in spine bone mineral density (BMD) 4 and improvements in total hip BMD 6 and spine BMD 6.

Romosozumab demonstrates significant efficacy in reducing the incidence of new vertebral fractures at 12 months (p<0.001) and 24 months (p<0.001), as well as clinical fractures at 12 months (p=0.008) 18. It also significantly increases BMD at the lumbar spine, total hip, and femoral neck [16, 21].

Abaloparatide is associated with significant increases in mineralizing surface/bone surface (MS/BS) in the cancellous envelope 20, as well as significant increases in lumbar spine, total hip, and femoral neck BMD at 12 months 22.

Alendronate combinations show significant improvements in lumbar spine BMD at both 6 and 12 months 13 and significantly higher gains in total hip, femoral neck, and lumbar spine BMD compared to other cohorts 17. Alendronate with cholecalciferol was associated with a reduction in patients with serum 25-hydroxyvitamin D below deficiency levels (p<0.0001) and significant changes in Serum PTH 15.

Ibandronate is associated with high patient adherence rates (98.5%) and high scores in patient satisfaction surveys 11. Patients also showed a preference for once-monthly dosing over once-weekly dosing based on convenience of administration (p<0.0001) 9.

Placebo groups showed significant increases in lumbar spine BMD compared to other cohorts at 6 and 12 months 12, while a separate placebo study showed significantly lower rates of new vertebral fractures compared to an intervention group 19.
