What the trials found Updated — new results For clinicians

Prostate adenocarcinoma: what the trials found

Short-term androgen deprivation has been shown to significantly improve clinical outcomes in prostate adenocarcinoma, with a 90% vs 81% rate of biochemical disease-free survival at 5 years (p=0.01), an 83% vs 94% metastasis-free survival rate (p=0.01), and an 86% vs 95% overall survival rate at 5 years (p=0.009) 5.

Imaging modalities including 68Ga-RM2 6, 68Ga-PSMA-11 7, Ga-PSMA-11 9, C-11 choline PET tracer 11, and 18F-DCFPyL have been evaluated in clinical trials. Notably, 131 participants had their intended treatment plans changed based on 18F-DCFPyL PET/CT imaging results 10.

Self-administered nitrous oxide was evaluated in a Phase 4 trial 4. Additionally, hypofractionated radiation therapy did not show statistically significant differences in the urinary or bowel domains of the Expanded Prostate Cancer Index (EPIC) at two years 8.

Recent results — preliminary, needs further review

  • Niraparib Pill showed a 6-month radiographic progression-free survival (rPFS6) of 6 (p=0.006), a PSA50 response in 3 patients (p=0.05), and a time to PSA progression of 11.2 (p=0.05) 2.
  • Androgen Suppression was evaluated in a Phase 2 trial 1.
  • Gonadotrophin Releasing Hormone was evaluated in a Phase 2 trial 12.

For the clinician treating this condition

  • Short-term androgen deprivation is associated with significant improvements in biochemical disease-free survival, metastasis-free survival, and overall survival at the 5-year mark.
  • Multiple PET tracers (68Ga-RM2, 68Ga-PSMA-11, Ga-PSMA-11, C-11 choline, and 18F-DCFPyL) have been established in clinical trials for imaging purposes.
  • Hypofractionated radiation therapy did not result in statistically significant changes to urinary or bowel EPIC scores at the two-year follow-up.
