Non-Hodgkin's Lymphoma: what the trials found

Brentuximab vedotin has demonstrated significant efficacy in specific subsets of Non-Hodgkin's Lymphoma. In a Phase 3 trial, it showed an Objective Response Rate (ORR4) of 54.7% compared to 12.5% (p<0.001), with a corresponding increase in Complete Response (CR) rates of 17.2% vs 1.6% (p=0.0002) and Progression-Free Survival (PFS) of 16.7% vs 3.5% (p<0.001) 17. Additionally, a Phase 4 study confirmed an ORR of 60.0% for patients with CD30+ MF or pcALCL 10.

Ibrutinib showed significant improvements in Progression-Free Survival (PFS). In one trial, the Kaplan-Meier estimate at month 30 was 78.5% vs 31.1% (p<0.0001), and at month 48, it remained significantly higher at 74.0% vs 22.0% (p<0.0001). Notably, in a high-risk sub-population (del17p/TP53 mutation/Del 11q), the PFS was 82.4% vs 14.1% (p<0.0001) 18.

Obinutuzumab demonstrated significant improvements in Progression-Free Survival (PFS) as assessed by both Independent Review Committee (14.1% vs 29.2%, p<0.0001) and Investigator assessment (14.1% vs 25.8%, p<0.0001) 20.

Other established interventions include rituximab, which showed a progression-free survival rate of 24.0% 14 and was utilized in autologous transplantation protocols with high rates of platelet recovery (over 80%) 21. Bexarotene showed varied results for tumor response duration (99.4% vs 54.1%) and time to response (66.1% vs 117.0%) 12. Temsirolimus was associated with specific rates of infection-related and bleeding-related adverse events 13.
