Spondyloarthritis: what the trials found

Several biologics and targeted synthetic DMARDs have demonstrated efficacy in treating spondyloarthritis. Ixekizumab showed significant improvements in clinical outcomes, with participants achieving ASAS40 responses (up to 51.8% in some cohorts) and ASAS20 responses ranging from 29.8% to 68.7% [4, 6]. Specifically, Ixekizumab demonstrated significant reductions in Ankylosing Spondylitis Disease Activity Score (ASDAS) compared to baseline [4, 6] and showed high rates of ASAS20 response in bDMARD-naïve patients 7.

Upadacitinib demonstrated statistically significant improvements in disease activity at week 14, with ASAS40 response rates reaching up to 44.9% and substantial reductions in ASDAS scores compared to baseline 3. Golimumab was associated with high percentages of participants remaining without a disease activity flare during Period 2 (up to 84.1%) and showed high clinical response rates following open-label retreatment for those who experienced flares 1.

Other interventions showed specific improvements in pain and index scores. AIN457 demonstrated significant results in reducing spinal pain (NRS score below 4) and improving Bath Ankylosing Spondylitis Disease Activity Index scores at week 8 5. Additionally, Adalimumab was associated with a reduction in the Lewis Score Index 2.

Recent results — preliminary, needs further review
