Extensive-Stage Small-Cell Lung Cancer: what the trials found

HLX10 has demonstrated statistically significant improvements in both overall survival (OS) and progression-free survival (PFS) for patients with extensive-stage small-cell lung cancer 1. Specifically, OS was recorded at 15.77 versus 11.10 months (p<0.001), while PFS reached 5.82 versus 4.34 months (p=0.001) 1.

Durvalumab treatment in this patient population showed that 15% of participants remained alive and progression-free at 12 months (PFS12) 2. Additionally, 19.8% of patients remained in response 12 months after their first documented objective response (DoR12), with 91 patients experiencing Grade 3 or higher adverse events 2.

The addition of Trilaciclib to carboplatin and etoposide, or Topotecan, significantly reduced the duration of severe neutropenia in Cycle 1 compared to other regimens (0.7 vs 0; 0.7 vs 2; p=0.0003) 3. This regimen also showed lower rates of composite endpoints for major hematologic adverse events, including all-cause hospitalizations and dose reductions, with reported rates of 0.033, 0.040, and 0.077 respectively 3.
